Background: The SPPLs are a group of aspartic proteases that are responsible for proteolysis of type II membrane-anchored signalling factors. SPPL2b, unlike other SPPL family members is found in endosomes/lysosomes, and not the endoplasmic reticulum, and has a role in catalysing intramembrane cleavage of tumour necrosis factor alpha (TNFalpha).
